How Liposomes can increase the solubility of an Insoluble Drug
Why Is Intravenous Administration So Important Pre-Clinically?
There are a number of reasons why IV administration is so critical to your poorly soluble compound at the Pre-Clinical stage:
- Absolute bioavailability: this informs you how much of your insoluble drug is systemically available after injection of the compound
- Critical pharmacokinetic data: this is vital to understanding your insoluble compound’s behavior in vivo
- Toxicity: see potentially tox effects in vivo after systemic administration
- Potency: essential to knowing the fundamental efficacy of your poorly soluble compound
- Relative bioavailability: how much of an oral dose is absorbed compared to an intravenous one
Liposomes are spherical structures which are very similar to naturally occurring cells in mammals. This carrier can be exploited to fit a poorly soluble compound into the liposome in a similar way to when a compound enters a cell membrane of the body.
Liposome Solubilizer is a handy tool designed specifically to solubilize poorly soluble drugs for Pre-Clinical intravenous administration. This enables you to establish absolute bioavailability (i.e. after intravenous delivery) of your insoluble compounds or to explore in vivo efficacy without drug precipitation.
Key Applications
The main applications for using Liposome Solubilizer with poorly soluble compounds are:
- Significantly increase the aqueous solubility of an insoluble drug
- Keep your insoluble compound in solution so it does not precipitate and cause blockage in the bloodstream (emboli- see above pic) or cause irritancy upon dilution or injection
- Avoid the use of surfactants such as Cremophor (see above pic) that can cause anaphylaxis and a high solvent level that‘s poorly tolerated
