Why Is Solubilization So Important?


If you have a poorly soluble compound intended for intravenous administration, you must understand how it will perform after injection. You need a solubilizer that not only dissolves your compound but also keeps it in solution. If it precipitates out after injection this could result in the physical blockage of capillaries (emboli). Worse still, promising activity could be missed because the drug is not free to act. Unfortunately, only a few existing solubilization techniques are suitable for intravenous administration. Many solubilizers are poorly tolerated in Pre-Clinical species. Solubilizing an insoluble drug is no easy task.


How Liposomes Can Help


Historically, liposomes have mostly been used for targeting water soluble compounds. It’s notoriously difficult to load in poorly soluble drugs with conventional approaches, so their role as a well-tolerated solubilizer has largely been overlooked. But liposomes can be good solubilizers because they can help prevent drug precipitation. Also, unlike Cremophor for example, drug-loaded liposomes can be used in Pre-Clinical models which are intolerant to such surfactants.
